ABSTRACT

Objective:To explore the application of lecture-based learning (LBL) combined with problem-based learning (PBL) in the teaching of pathophysiology case analysis.Methods:The subjects were 62 preventive medicine majors of Grade 2015 and 68 stomatology majors of Grade 2015, and the course content was case analysis. PBL teaching was implemented in the preventive medicine major group and LBL+PBL teaching was implemented in the stomatology major group. Both groups received 6 class hours, among which the LBL+PBL teaching hours were allocated 2 class hours for LBL and 4 class hours for PBL. In addition, the questionnaire was used to evaluate the teaching effects between the two groups, and the descriptive analysis was used to evaluate relevant data lines.Results:Eleven students (17.742%) from the preventive medicine major group chose PBL teaching, while 38 students (92.647%) in the stomatology major group held a very positive attitude towards LBL+PBL teaching. It can be seen that the recognition degree of LBL+PBL teaching in the stomatology major group was higher than that of PBL teaching in the preventive medicine major group.Conclusion:Since students have received LBL teaching mode for a long time, it is difficult to accept and adapt to PBL teaching in terms of thinking and teamwork. Therefore, in the course of pathophysiology case analysis, LBL+PBL teaching shows a good effect, which is worthy of further exploration.

ABSTRACT

Objective:To observe the morphological features of liver injury induced by hydrodynamic injection, and to explore the procedure and mechanism of liver repair. Methods:Twenty-five female Balb/c mice were intravenously injected with large volume of saline solution(1.8-2.0 mL)in 3 s. The mice were divided into 30 min,8 h,1 d,3 d,and 7 d groups according to the post-injection time.Each group contained 5 mice.Six non-injected mice were used as control group.The blood samples from angular vein of the mice were collected to detect the levels of alanine transaminase(ALT).The morphological features of liver tissue of the mice in various groups were observed with HE staining.The proliferating cell nuclear antigen(PCNA)protein expressions in liver tissue of the mice in various groups were detected by immunohistochemical saining.The expression levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),epidermal growth factor(EGF),hepatocyte growth factor (HGF),transforming growth factor-α(TGF-α),Cyclin D1,vascular endothelial growth factor(VEGF),Bax and Bcl-2 mRNA were determined by Real-time PCR.Results:Compared with control group,the ratio of liver weight/original body weight in 8 h group was significantly decreased(P<0.05),but there was no significant difference in 7 d group(P>0.05).Compared with control group,the serum ALT level in 1 d group was significantly elevated (P<0.01),but there was no significant difference in 7 d group(P>0.05).Compared with control group,the liver tissue of the mice in 30 min group showed hepatocyte swollen and small hemorrhagic area,and the number of hepatocytes per high power field was significantly declined(q=4.760,P<0.05)under microscope.Compared with 30 min group,the number of swollen cells was decreased in 8 h group,the hemorrhagic and necrotic area enlarged, both of the hepatocyte number and binuclear hepatocyte number per high power field were significantly increased (q=7.310,P<0.01;q=7.200,P<0.01).Compared with 8 h group,the hemorrhagic and necrotic areas in 1 d group were reduced,both of the hepatocyte number and binuclear hepatocyte number per high power field were significantly decreased(q=4.966,P<0.05;q=6.596,P<0.01);there were no significant differences compared with control group(P>0.05).The hemorrhagic and necrotic areas almost disappeared in 3 d group.The histological appearance of liver tissue of the mice in 7 d group was same as control group.Compared with control group,the PCNA indexes in hepatocytes in 8 h and 1 d groups were significantly increased(t=4.458,P<0.01;t=15.557,P<0.01);there was no significant difference in 7 d group(P>0.05).Compared with control group, the PCNA index in cholangiocytes in 30 min group was increased significantly(t=3.985,P<0.01);there was no significant difference in 7 d group(P>0.05).The mRNA expression levels of TNF-α,IL-6,EGF,and VEGF in liver tissue of the mice in 30 min group were significantly higher than those in 8 h group(q=4.952,P<0.05;q=14.750,P<0.01;q=14.750,P<0.01;q=13.551,P<0.01).The HGF mRNA expression level in liver tissue of the mice in 3 d group was significantly higher than those in 30 min group and 8 h group(q=5.031,P<0.05;q=4.631,P<0.05).The TGF-αmRNA expression levels in liver tissue of the mice in 8 h group and 1 d group were higher than that in 30 min group(q=4.592,P<0.05;q=8.137,P<0.01).The Cyclin D1 mRNA expression levels in liver tissue of the mice in 8 h group and 1 d group were higher than that in 7 d group(q=4.736, q=5.213,P<0.05).The Bax/Bcl-2 ratio of the mice in 1 d group was higher than that in 30 min group(q=5.731,P<0.01).Conclusion:Acute liver injury induced by hydrodynamic injection mainly shows the hepatocyte swelling and hemorrhagic necrosis.The injury could be repaired naturally in a week.The various cytokines and growth factors related with liver regeneration are involved in the repair procedure.

ABSTRACT

AIM:To explore the effects of pGRIM-19-si-survivin co-expression plasmid carried by human at-tenuated Salmonella on prostate cancer subcutaneous xenograft growth in nude mice .METHODS: Prostate cancer xeno-graft model was established in nude mice .Co-expression plasmids carried by attenuated Salmonella were introduced by in-traperitoneal injection .The xenograft volumes were monitored timely .Immunohistochemical staining , RT-PCR and TUNEL assay were applied to investigate the related mechanisms that pGRIM-19-si-survivin inhibited tumor growth in vivo.RE-SULTS:Compared with psi-survivin and pGRIM-19 carried by attenuated Salmonella ( control groups ) , the tumor volumes were reduced markedly in pGRIM-19-si-survivin plasmid group .The mean shrinkage rates were 2.36 and 3.02 times. pGRIM-19-si-survivin co-expression plasmid carried by attenuated Salmonella inhibited survivin expression but strengthened GRIM-19 expression obviously (P<0.05).The mRNA expression of apoptosis-related proteins such as Bcl-xL, Stat3, cy-clin D1 and c-Myc was inhibited , and the vascular endothelial growth factor ( VEGF) mRNA and Ki67 protein were also in-hibited, but the caspase-3 mRNA expression was up-regulated ( P<0.05 ) with significant cell apoptosis .CONCLU-SION:pGRIM-19-si-survivin co-expression plasmid carried by human attenuated Salmonella inhibits the growth of prostate cancer subcutaneous xenografts in nude mice by promoting cell apoptosis and inhibiting prostatic cancer proliferation .

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the expressions of survivin and GRIM-19 in prostatic cancer tissue and their clinical implications.</p><p><b>METHODS</b>We detected the expressions of survivin and GRIM-19 in the tissues of normal prostate (NP), benign prostate hyperplasia (BPH) and prostate cancer (PCa) using immunohistochemical staining, RT-PCR and Western blot, and processed the data by SPSS12.</p><p><b>RESULTS</b>The positive rates of survivin expression were 6.25% , 18.18% and 90.62% in NP, BPH and PCa (P < 0.01), while those of GRIM-19 were 87.50%, 81.82% and 9.37% , respectively (P < 0.01). Semiquantitative RT-PCR and immunohistochemical staining showed that both survivin mRNA and survivin expressions were highly positive in PCa but negative in NP and BPH. Western blot exhibited that the survivin protein was expressed strongly in PCa but weakly in NP and BPH, while the GRIM-19 protein was expressed just contrariwise (P < 0.01).</p><p><b>CONCLUSION</b>The expressions of survivin and GRIM-19 may be closely correlated with the pathogenesis of prostate cancer.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the therapeutic effect of catgut implantation at acupoint on simple obesity of different syndrome types, so as to understand the syndrome type most suitable to this therapy.</p><p><b>METHODS</b>Using prospective and double blind method one hundred and thirty-seven cases were divided into a group of dampness stagnancy due to spleen deficiency (n=30), a stomach heat-dampness stagnation group (n=30), a group of stagnation of liver qi (n=27), a group of deficiency of both spleen and kidney (n=26) and a group of endogenous heat due to yin deficiency (n=24). They were treated with catgut implantation at main acupoints Liangmen (ST 21), Zhongwan (CV 12), Tianshu (ST 25), Qihai (CV 6), Fenglong (ST 40) and adjuvant points selected according to different syndromes, once each week, 4 weeks constituting one course, for 2 courses. Changes of body weight, body mass index (BMI), circumferences of chest, waist and hip, and symptoms and signs before and after treatment were observed.</p><p><b>RESULTS</b>After treatment, the body weight, BMI, circumferences of waist and hip in the 5 groups had significant changes; the total effective rates in the group of dampness stagnancy due to spleen deficiency, the stomach heat-dampness stagnation group, the group of stagnation of liver qi, the group of deficiency of both spleen and kidney deficiency, the group of endogenous heat due to yin deficiency were 83.3%, 93.3%, 48.2%, 69.2% and 41.70%, respectively.</p><p><b>CONCLUSION</b>The therapeutic effect of catgut implantation at acupoints is the best for simple obesity of the stomach heat-dampness stagnation type and the worst for the type of endogenous heat due to yin deficiency.</p>

ABSTRACT

AIM: To study the protective effects of basic fibroblast growth factor (bFGF) on myocardial ischemia in rats and their underlying mechanism. METHODS: A rat myocardial ischemic injury model was established by left coronary artery ligation. The rats were killed at 2 h, 4 h, 8 h after coronary artery occlusion. The samples of blood and myocardium were collected for observing the expression of Bcl-2 and Bax in myocardial cells and the changes of superoxide dismutase (SOD) or myocardial enzymes.
